ACE abundance / catalytic activity / localization / neural circuits
The outcome of a single ACE inhibitor cannot determine ACE’s overall involvement or roles. The findings motivate tests separating abundance, catalytic activity and localization.
This cross-sectional association does not establish causal function or efficacy in depression. Earlier captopril findings also do not prove a nonenzymatic role for ACE.
Separate abundance, activity and localization in patient-derived cells and mice; distinguish circuit and behavioral measures from blood-pressure and locomotor effects.
What would change our view: Reconsider the hypothesis if separated manipulations do not reproduce predicted neural effects or associations are explained by confounding or peripheral effects.
A prospective test proposal, not a prediction score. A graph connection does not validate this hypothesis.

Reading at the time: The study reports lower ACE protein abundance associated with early-stage psychosis and treatment resistance, but no significant group difference in enzyme activity per unit serum volume.
